I think it can be, but I never tested that. And the result would be 'granted'.

Actually, i wanted to use cordova-plugin-ace and requesting permissions not defined in AndroidManifest.xml is very essential in this case.

First at all, I need to apologize that I didn't fully understand your requirement before.
You can not request any permission that not defined in AndroidManifest.xml. That is totally disobey this Android security design.
Those permissions that you declared on AndroidManifest.xml are just telling your app user that your app will use those permissions in the runtime. The goal of runtime requesting permissions is let you ask users do they want to grant those permissions to your app.
The conclusion - NO.
