SELinux most likely causes this issue. If you would like to try ec2-instance-connect on CentOS, in most cases, disabling SELinux is the fastest (and the most insecure) way.
As for your question, we do have plans for CentOS, RHEL, or other systems that have SELinux on by default, please take a look at #2 .

Disabling SELinux is undesirable - I will take a look at the guidance offered in #2 and report my results.
